ALTERNATIVE IN VITRO METHODS FOR THE EVALUATION OF THE TOXIC POTENTIAL OF CHIMICAL COMPOUNDS VALIDATED FROM EURL ECVAM (https://eurl-ecvam.jrc.ec.europa.eu)

 

 

Nowadays ECVAM has successfully validated at least 25 replacemnet, reduction and refinment methods, among which more than 15 already had been accepted at a regulatory level for the safety assessment of chemical compounds.

LARF offers:

 

In vitro models

  • established human normal cell lines of keratinocytes, endothelial cells, fibroblasts, etc (on customer request);
  • established human cancer cell lines from variuos organs (on customer request);
  • co-culture of human cell lines;
  • 3D models of organs reconstituted on dedicated supports (scaffold, hydrogel, biomatrices), on request.

 

In vitro analyses

 

In vitro evaluation of cellular damage related to:

  • cytotoxicity
  • cellular proliferation
  • anti/pro- oxidant potential
  • anti/pro-inflammatory activity
  • sensitizing potential
  • enzymatic activities of phase 1 and phase 2
  • gene and protein expression and localization of markers of the cellular damage, oxidative stress, irritation/inflammation, cellular activation.

 

Our staff performs techniques of molecular biology for the study of the expression of cellular markers involved in cellular damage, in skin aging, in the activation of the response to inflammatory stimuli and oxidative stress, etc.

 

On customer request, we perform analyses of proteins of interest and their possible modulation after exposure to the chosen cell models, normal and/or cancer, at the chemical compound under examination.

 

We also analyse the levels of:

  • Protein expression by immunoblotting and/or ELISA
  • mRNA expression by RT-PCR

 

Subcellular localization (immunofluorescence/immunohistochemistry).

•Mid-throughput electrophysiological recording of in vitro neuronal cultures coupled to microelectrode array (MEA).
